產(chǎn)品介紹 簡介
蛋白質(zhì)定量是蛋白質(zhì)研究的基礎(chǔ)工作之一。BCA (bicinchoninic acid)是理想的蛋白質(zhì)定量方法;是當(dāng)前比Lowry法更優(yōu)越的專用于檢測總蛋白質(zhì)含量的產(chǎn)品。該方法以快速靈敏、穩(wěn)定可靠,對不同種類蛋白質(zhì)檢測的變異系數(shù)非常小而倍受專業(yè)人士的青睞。BCA法測定蛋白濃度不受絕大部分樣品中的化學(xué)物質(zhì)的影響。在組織細(xì)胞裂解實(shí)驗(yàn)中,常用濃度的去垢劑SDS,Triton X-100,Tween不影響檢測結(jié)果,但受螯合劑(EDTA,EGTA)、還原劑(DTT,巰基乙醇)和脂類的影響。實(shí)驗(yàn)中,若發(fā)現(xiàn)樣品稀釋液或裂解液本身背景值較高,可試用Bradford蛋白濃度測定試劑盒。該產(chǎn)品不受此影響,因此與之配合使用,可免除您實(shí)驗(yàn)中的許多煩惱。
基本原理:堿性條件下,蛋白將Cu2+還原為Cu+, Cu+與BCA試劑形成紫顏色的絡(luò)合物,測定其在562nm處的吸收值,并與標(biāo)準(zhǔn)曲線對比,即可計算待測蛋白的濃度。
BCA試劑的蛋白質(zhì)測定范圍是10-2000ug/ml

使用說明:
1. 配制工作液: 根據(jù)標(biāo)準(zhǔn)品和樣品數(shù)量,按體積比A :B (50:1)配制適量BCA工作液,充分混勻。BCA工作液室溫24小時內(nèi)穩(wěn)定。
2. 稀釋標(biāo)準(zhǔn)品:取10微升標(biāo)準(zhǔn)品用PBS稀釋至100ul(標(biāo)準(zhǔn)品一般可用PBS稀釋),使終濃度為0.5mg/ml。將標(biāo)準(zhǔn)品按0, 1, 2, 4, 8, 12, 16, 20ul加到96孔板的蛋白標(biāo)準(zhǔn)品孔中,加PBS補(bǔ)足到20ul。
3. 加適當(dāng)體積樣品到96孔板的樣品孔中,補(bǔ)加PBS到20ul。
4. 各孔加入200ul BCA工作液,37℃放置30分鐘。
5. 冷卻到室溫,用酶標(biāo)儀562nm測定OD值,根據(jù)標(biāo)準(zhǔn)曲線計算出蛋白濃度。

注意事項(xiàng):
1. 在低溫條件或長期保存出現(xiàn)沉淀時,可攪拌或37℃溫育使溶解, 如發(fā)現(xiàn)細(xì)菌污染則應(yīng)丟棄。
2. 樣品中若含有EDTA、EGTA、DTT、硫酸銨、脂類會影響檢測結(jié)果,請試用Bradford蛋白濃度測定試劑盒;高濃度的去垢劑也影響實(shí)驗(yàn)結(jié)果,可用TCA沉淀去除干擾物質(zhì)。
3.要得到更為精確的蛋白濃度結(jié)果,每個蛋白梯度和樣品均需做復(fù)孔, 每次均應(yīng)做標(biāo)準(zhǔn)曲線。
4.當(dāng)試劑A和B混合時可能會有渾濁,但混勻后就會消失,工作液在密閉情況下可保存1周。
5.需準(zhǔn)備37℃水浴或溫箱、酶標(biāo)儀或普通分光光度計,測定波長為540-595nm之間,562nm最佳。酶標(biāo)儀需與96孔酶標(biāo)板配套使用。使用分光光度計測定蛋白濃度時,試劑盒測定的樣品數(shù)量會因此而減 少。使用溫箱孵育時,應(yīng)注意防止因水粉蒸發(fā)影響檢測結(jié)果。
